RNA-seq of major tissues and xylem cell types of Populus trichocarpa We used the stem cross-sections of P. trichocarpa and our recently developed laser capture microdissection (LCM) to collect fibers, vessels, and a combination of 3 cell types (fibers + vessels + rays). Total RNA from the three samples was isolated, amplified, and analyzed by full-transcriptome RNA-sequencing. The sequencing reads were mapped to the P. trichocarpa genome. Overall design: Different major tissues (shoot tip, leaf, phloem, primary root, stem differentiating xylem) and xylem tissue c......[More] Forest Biotechnology Group, Forestry and Environmental Resources, North Carolina State University 8
Ptr-miR397a is a negative regulator of laccase genes affecting lignin content in Populus trichocarpa Laccases were proposed to catalyze the oxidative polymerization of monolignols. We identified 49 laccase gene models in Populus trichocarpa, of which 29 were predicted to be targets of ptr-miR397a. We overexpressed Ptr-MIR397a in transgenic P. trichocarpa. In each of all 9 transgenic lines tested, 17 PtrLACs were down-regulated as analyzed by RNA-seq. Transgenic lines with severe reduction in the expression of these laccase genes resulted in an approximately 40% decrease in the total laccase act......[More] Forest Biotechnology Group, Department of Forestry and Environmental Resources, North Carolina State University 2
P. trichocarpa responds to drought stress at physiological and transcriptome levels Our analysis provides a comprehensive picture of how P. trichocarpa responds to drought stress at physiological and transcriptome levels which may help to understand molecular mechanisms associated with drought response and could be useful for genetic engineering of woody plants. SNP discovery in black cottonwood (Populus trichocarpa) by population transcriptome resequencing. IN THIS POPULATION BASED TRANSCRIPTOME ANALYSIS WHOLE MRNA WAS EXTRACTED FROM DEVELOPING XYLEM OF 20 P. TRICHOCARPA NATURAL ACESSIONS AND SEQUENCED WITH ILLUMINA%u2019S GA II ANALYZER AFTER CONVERSION TO CDNA AND PREPARATION OF PAIRED END LIBRARIES. THESE DATA WERE MAPPED TO THE REFERENCE GENOME OF P. TRICHOCARPA (version 2) AND USED TO DISCOVER SNPS IN GENES THAT ARE EXPRESSED DURING WOOD FORMATION. University of British Columbia 20
