Description

The TIFY family constitutes a plant-specific group of protein with a broad range of functions. This family encodes four subfamilies of proteins, including ZML, TIFY, PPD and JASMONATE ZIM-Domain (JAZ) proteins. This family owes their name to a conserved motif (TIF[F/Y]XG) located within an approximately 36 amino acid long TIFY domain. While all TIFY proteins bear a TIFY domain, those in the ZML subfamily, also contain both a C2C2-GATA zinc-finger DNA-binding domain and a CCT domain (CONSTANS, CO-like, TOC1). Conversely, proteins from both PPD and JAZ subfamilies lack GATA and CCT domains. Interestingly, in addition to the TIFY domain, the JAZ subfamily also contain a conserved sequence of approximately 27 amino acids near their C-terminus, referred to as the Jas motif, which is similar in sequence to the N-terminal portion of the CCT domain and bears the characteristic motif SLX2FX2KRX2RX5PY. PPD proteins, on the other hand, bear a unique N-terminal PPD domain, as well as a divergent Jas motif that lacks the conserved PY at its C-terminus. Finally, proteins from the TIFY subfamily contain only the TIFY domain.
